对8株茶卡盐湖附近土壤分离乳杆菌进行益生性的检测,以期获得具有较高益生活性的乳酸菌。采用纸片扩散法、人工模拟胃肠道环境来检测乳杆菌对抗生素的药敏性以及对胃肠道环境的耐受性。结果显示,8株菌株对万古霉素、庆大霉素、多黏菌素B、环丙沙星、青霉素G具有抗性,对氯霉素、氨苄西林、四环素较为敏感。在p H为1~2的极端酸性环境中,乳杆菌的生长受到抑制,几乎不能生存,但对p H为3的酸性环境耐受性较强。通过人工模拟胃肠道环境耐受性的检测,发现L.plantarum N20、L.pentose N7、L.pentose N6耐受性较好,存活率依次为61.11%、51.25%、50.00%。胆盐对乳杆菌的生长显示出抑制性,并且随着胆盐浓度的增加,抑制活性逐渐增强。其中L.plantarum N22、L.plantarum N19在0.3%的胆盐浓度下4 h活菌数仍有5.00×102,具有一定强度的胆盐耐受性。
The probiotic properties of eight Lactobacillus isolated from soil of Caka Salt Lake area were analyzed.The susceptibility to 10 kinds of commonly used antibiotics, gastrointestinal environment, bile and p H tolerability were detected using diffusion technique in an agar and simulated artificial gastrointestinal environment.The results showed that all of them were resistant to vantamicin,polymyxin B,ciprofloxacin and penicillin G but sensitive to chloramphenicol,ampicillin,and tetracycline. In the extreme acidic environment,for example,p H was 1 or 2,the growth of lactobacillus could not survive,but when the p H was 3,they could grow. The results indicated that L.plantarum N20,L.pentose N7,L.pentose N6 exhibited better tolerability in the test of artificial gastrointestinal environment simulation,and survival rate were 61.11%,51.25%,and 50% respectively.Bile salts showed inhibitory influence on the growth of lactobacillus;with the increase of bile salt concentration,the inhibition activity gradually enhanced. The number of L.plantarum N22,and L.plantarum N19 was 5.00 ×102at 0.3% bile salt for 4 h indicating a good bile salt tolerance.
