[目的]探讨Aurora—A基因在不同组织类型肺癌中的表达及其与临床的相关性。[方法]采用RT—PCR、Western—blot与免疫组化方法检测肺癌组织标本行Aurora—A的mRNA与蛋白表达。[结果]肺癌30例标本中RT—PCR检测Aurora—A mRNA的表达,19例(63%)肺癌表达水平高于相应癌旁组织,其中Westernblot方法检测20例中,13例(65%)蛋白表达水平高于相应癌旁组织。免疫组化检测48例肺癌中32例高表达(66.67%)。肺癌Aurora—A mRNA的表达水平与肺癌患者的性别,吸烟史以及病理特征无明显相关性。肺鳞癌的Aurora—A的蛋白水平高表达程度与肿瘤转移有相关性。[结论]肺癌组织Aurora—A表达明显高于其癌旁组织,提示Aurora—A基因的过表达与肺癌的发生有相关性。
[ Objective] To study Aurora - A gene expression status of lung cancer and its influence on genesis of lung cancer in vivo. [ Methods ] The levels of Aurora - A mRNA and protein of lung cancer samples were detected by RT PCR, Westem Blot or/and immunohistochemistry respectively. [ Results ] In 30 cases of lung cancer patients, the over - expressions of Aurora - A mRNA were observed in 19 cases (63%) compared with neighboring tissues (P 〈 0.05). Westem blot was adopted to assess the status of Aurora - A protein expressed in tumor in vivo from 20 patients. The over - expressioof Aurora - A protein n was detected in 13 cases (65%) compared with their neighboring tissue(P 〈0.05). Correspondingly, over expression were also found in 32 cases (66.67%) of 48 cases dectected by immunohistochemistry. No significant correlation existing between over expression of Aurora - A mRNA and sex, smoking history and pathological characteristics was found. On the contrary, correlation was observed between over expression level of Aurora - A protein and metastases in 48 squamous lung cancer patients. [ Conclusion] The Aurora -A is highly expressed in lung cancer tissue than in neighboring tissue, suggesting the over expressed Aurora - A be one of oncogenetic factors of lung cancer.
