目的优化云南建水紫陶土耐酸真菌Penicillium purpurogenum JS03-21产肿瘤细胞毒活性的purpurquinoneA的发酵条件,提高purpurquinoneA的产量。方法采用单因素实验,对培养方式、培养时间、pH值、pH调节方式、温度、摇床转速、装液量、种龄、接种量、碳源、氮源、无机盐等条件进行研究,通过正交试验,确定最佳培养基组成。结果PurpurquinoneA最佳发酵条件为:装液量150/500mL、种龄4d,接种量为4%,28℃、180r/min下摇床发酵15d;最佳培养基组成为:50g麦芽糖、13g牛肉膏、2gMgSO4、1000mL自来水、10%盐酸调节初始pH2.0。结论以最佳发酵条件发酵,优化后purpurquinoneA产量达到547mg/L,是优化前产量的96倍。
Objective To optimize the fermentation conditions and to improve the yield of the purpurquinone A, a cytotoxic azaphilone from a purple clay-derived Penicillium purpurogenum JS03-21 collected from Jianshui, Yunnan, China. Method Using single-factor experiments to study the culture way, the cultivation time, pH, temperature, the shaking speed, carbon sources, nitrogen sources, inorganic salts, inoculum size and pH adjustment mode. And the optimized composition of the medium was investigated by orthogonal matrix methods. Results The optimized fermentation conditions are 150 mL of culture medium in 500-mL conical flask, 4 days old of inoculum age and 4% of inoculum size, and shaking in an orbital shaker (180 r/min) for 15 days at 28℃. The composition of the optimized medium is 50g maltose, 13g beef extract, 2g MgSO4, 1000mL tap-water and adjusting the initial pH value to 2.0 by 10% HCl. Conclusion Compared to the original condition, the best yield of purpurquinone A reached 547mg/L with a 96-fold increase under the optimized fermentation conditions.
