目的:研究葛根素与葛根芩连汤主要有效成分配伍时各有效成分在肠道中的吸收动力学变化,探讨复方配伍的规律性。方法:通过Caco-2细胞单层模型及超高效液相色谱法(UPLC)研究方中有效成分葛根素与黄芩苷、盐酸小檗碱、甘草苷进行配伍时,各有效成分的肠道吸收变化情况。结果:以各有效成分的表观渗透系数(Papp)及吸收率(%)为指标,得出同一成分在不同配伍组中肠道吸收情况有所不同,并且葛根素的肠道吸收情况在各配伍组具体为葛根素-黄芩苷-小檗碱-甘草苷配伍组〉葛根素.黄芩苷配伍组〉葛根素-小檗碱配伍组〉葛根素-甘草苷配伍组〉单独葛根素(P〈0.05)。结论:葛根芩连汤有效成分在肠道中吸收动力学过程随方中有效成分的配伍情况不同而变化,方中其他药味成分对葛根素的肠道吸收具有促进作用。
Objective:To study the intestinal dynamics characteristics of puerarin and other active ingredients in Gegenqinlian de- coction. Methods:Caco-2 cell monolayer model was used to research the transport of puerarin and baicalin, berberine hydrochloride, liquiritin when they were used in different compatibilities, meanwhile a ultra performance liquid chromato-gl:aphic method was also de- veloped. Results :The concentration of puerarin, baicalin, berberine hydrochloride, liquiritin in cell culture medium was measured and the apparent permeability coefficients ( Papp ) as well as percent absorption ( % ) from apical side to basolateral side were calculat- ed. The absorption characteristics of the four components were different when they were used in different eompatibilities. Furthermore the P,pp sequence of puerarin in specific groups were puerarin-baicalin-berberine-liquiritin group 〉 puerarin-baicalin group 〉 puerarin-ber- berine 〉 puerarin-liquiritin group 〉 single puerarin ( P 〈 O. 05 ). Conclusion : The intestinal absorption dynamics process of the active in- gredients in Gegenqinlian decoction changes as the change of prescription compatibilities. And the intestinal absorption characteristics of puerarin is enhanced by the use of other components.
