目的采用大鼠肠外翻模型研究葛根芩连汤不同配伍对黄连中生物碱的肠吸收特性。方法以大鼠肠道P-糖蛋白(P-gp)抑制剂维拉帕米为判断依据,比较葛根芩连汤中黄芩和葛根对黄连生物碱吸收参数Ka。结果在维拉帕米的作用下,各生物碱在各肠段的吸收均有显著性增加(P〈0.05);葛根在十二指肠、空肠段可以使各生物碱成分的吸收增加(P〈0.05);黄芩在回肠和结肠段能使黄连碱、小檗碱和巴马汀成分的吸收增加(P〈0.05);而配伍甘草后基本无显著性增加。结论研究表明,葛根、黄芩可能通过对P-gp抑制,可以显著增加黄连生物碱类成分的吸收。
AIM To study the effect of Gegen Qinlian Decoction on rat intestinal absorption behavior of Coptidis Rhizoma alkaloids through everted gut sacs in vitro.METHODS Rat intestinal P-glycoprotein(P-gp) efflux pump inhibitor verapamil was a good predictor for the comparison of absorption parameter between Puerariae lobatae Radix-Coptidis Rhizoma and Scutellariae Radix-Coptidis Rhizoma.RESULTS The research indicated that under the action of verapamil,Ka value of Coptidis Rhizoma alkaloids markedly increased in four segments(P〈0.05) and increased in duodenum,jejunum segments for Puerariae lobatae Radix(P〈0.05).The Ka values of coptisine,berberine and palmatine were significantly different in colon and ileum segments for Scutellariae Radix(P〈0.05),while had no significant changes for Glycyrrhizae Radix et Rhizoma praeparata cum melle.CONCLUSION The experiment shows that Puerariae lobatae Radix and Scutellariae Radix play a P-gp inhibitor's role in promoting the intestinal absorption of Coptidis Rhizoma alkaloids.
