初步研究了番茄未成熟花粉离体成熟培养的可能性.从番茄花中分离出的双核早期花粉用&液体培养基离体培养7~9d后,有1%的未成熟花粉培养成熟,并萌发出花粉管.同时,对不同时期花粉蛋白表达做了SDS-PAGE分析,结果:番茄花粉在单核期出现了分子量为21.2kD,26.0kD,33.4kD,69.1kD和97.1kD的5种特异蛋白,双核期出现了分子量为11.7kD,16.9kD,37.3kD,41.2kD和47.1kD的5种特异蛋白,萌发花粉管后出现了分子量为20.5kD,82.8kD和105.3kD的3种特异蛋白.随着花粉的发育,其蛋白质格局也在不断发生变化,单核期和双核期花粉中相对分子量小的蛋白质明显较多.
The possibility of in vitro maturation of tomato immature pollen was studied. After pollen at early binucleate stage isolated from tomato flowers had been in vitro cultured in the liquid medium B3 for 7-9 days, 196 immature pollen maturated and germinated pollen tubes. At the same time, proteins extracted from pollen at different developmental stages were separated by SDS-PAGE. The electrophoresis spectrums indicated: Pollen at uninueleate stage appeared five special proteins which the molecular weights were 21.2 kD, 26.0 kD, 33.4 kD,69.1 kD and 97.1 kD. The proteins of 11.7 kD, 16.9 kD, 37.3 kD, 41.2 kD and 47.1 kD appeared in pollen at binueleate stage. Three proteins of 20.5 kD,82.8 kD and 105.3 kD were expressed at the stage of germination of pollen. Protein-configuration varied as pollen developed. Many small- molecular weight proteins were expressed in pollen at uninueleate and binueleate stages.
