本研究克隆出了麻疯树亚硝基谷胱甘肽还原酶(JcGSNOR)cDNA 的全长序列,并分析了其在麻疯树各部位中的表达情况.结果表明,在所有参试的器官中,JcGSNOR基因皆有表达,其中在种子种表达量最大,其次为根茎,叶表达量最少.对新基因编码蛋白的理化性质进行分析后发现JcGSNOR的分子量约为40.256 kDa ,等电点约为6.74;同源性分析表明,JcG-SNOR基因与其它植物 GSNOR基因核甘酸序列相似度达到80%以上,说明JcGSNOR是GSNOR蛋白家族的一个新发现的成员.同时将JcGSNOR基因与pYES2表达载体相连接后,克隆进酿酒酵母,并经RT-PCR验证后证实得到JcGSNOR的转基因酵母.利用GSNO敏感性实验分析了JcGSNOR的酶学活性.并在GSNO处理条件下,检测了转基因酵母和野生型的生长曲线.结果表明,JcGSNOR对酵母的生长没有明显影响,但能明显提高酵母中GSNOR的活性.
JcGSNOR was cloned by RT-PCR and RACE techniques from Jatropha curcas .The expres-sion situation of JcGSNOR in different organs of Jatropha curcas was detected by real time PCR .The result showed that ,JcGSNOR was expressed in all tissue detected .The expression level was the highest in the seed ,root and stem were more or less ,and leaf was the least .With bioinformatic analysis ,the JcGSNOR may be comprised of 379 amino acids ,to give the molecular weight of 40 .256 kDa and the pI value of 6 .74;Result analyzed by NCBI showed that ,JcGSNOR has up to 80% similarity with GSNOR of other plants ,which indicated JcGSNOR is a new member of GSNOR family .To identify the biochemi-cal activity of JcGSNOR ,the gene encoding JcGSNOR was cloned into saccharomyces cerevisiae .GSN-OR activity and growth curve of yeast were detected .This study indicated that ,JcGSNOR showed no obvious effect on grow th of yeast ,but enhanced the GSNOR activity significantly .
