目的:建立测定齐多夫定含量的新方法。方法:以甲基丙烯酸为功能单体,三甲氧基丙基三甲基丙烯酸酯为交联剂,合成抗HIV药物齐多夫定分子印迹聚合物,将其用于固相萃取,通过渗漏实验选择功能单体并考察了最大吸附量和最佳上样条件。结果:当以浓度为4.5 mmol·L^-1,pH=7的缓冲溶液上样时,最大吸附量达到59.7 mg·g^-1。并通过对上样、洗脱条件进行优化,成功地将齐多夫定从其结构类似物拉米夫定的混合物中分离出来。不同齐多夫定和拉米夫定加入量的回收率分别为101.6%~102.6%,103.4%~104.0%,RSD分别为1.6%~4.2%,1.4%~3.3%。结论:该方法快速、直观,可用于药物的分离与富集。
Objective:To establish a new method for determination of zidovudine(AZT).Methods:Using methacrylic acid as functional monomer,and trimethlolpropane trimethacrylate as crosslinking agent,molecularly imprinted polymers(MIP) specific for anti - HIV drug zidovudine were synthesised and packed into the SPE cartriges.Functional monomer,the maximum binding capacity and the best loading conditions were optimised by breakthrough experiment. Result:When the sample loaded at the concentration of 4.5 mmol·L^-1 NaH2PO4/NaOH buffer(pH 7.0),the maximum adsorption capacity was up to 59.7 mg·g^-1 of polymers.The eluting condition was also optimized, zidovudine and its analog(lamivudine) were separated successfully by the MIP -SPE method.It was found that the proposed method can be used for the determination of AZT and 3TC in the mixture with good recoveries of 101.6%- 102.6%and 103.4%- 104.0%,RSDs are 1.6%-4.2%and 1.4%-3.3%.Conclution:The method is rapid and direct for separation and enrichment of drug.
