研究了二甲酚橙(XO)在乙醇-无机盐双水相萃取体系中的萃取行为,对无机盐种类、缓冲溶液和其用量等条件进行了优化,在最优条件下研究了上下层体积与萃取率、分配比等因素的变化规律。结果表明:在使用K2HPO4-KH2PO4缓冲溶液5.0 mL,95%乙醇6.0 mL,K2HPO4 6.0g,乙醇和蒸馏水体积比为1:2的条件下,获得了最大萃取率96.5%,回归方程y=0.0004+0.0263x,相关系数r=0.9995,摩尔吸光系数ε=1.9×104 L/(mol.cm),线性范围0~25μg/mL。回收率为96.3%~98.5%。
Xylenol orange was used to investigate the extraction behavior of aqueous two-phase system of ethanol and inorganic salt mixture.Experimental conditions were optimized including inorganic salt,buffer,and doses.Under optimized conditions,the ratio of upper and lower volume,the extraction rate and distribution ratio have been studied.The results showed that the maximum extraction rate of 96.5% was obtained under 5.0 mL of KH2PO4-K2HPO4 buffer solution,6.0 mL of 95% ethanol,6.0 g of K2HPO4 salt and the ethanol-water volume ratio of 1:2.The linear regression equation was y=0.0004+0.0263x with correlation coefficient of R=0.9995.The molar absorption coefficient was ε=1.9×104L/(mol.cm) and the liner range was 0~25μg/mL with recoveries of 96%~99%.
