目的 从分子水平研究大鼠肝癌模型及正常组织的代谢改变.方法 对二乙基亚硝胺(DEN)诱导的Wistar大鼠(实验组,50只)肝脏肿瘤组织及正常组15只进行14.1T离体高分辨魔角旋转磁共振波谱分析,通过波谱分析专用软件及一系列校正后,利用正交信号校正偏最小二乘法(OPLS),分析肝脏肿瘤的代谢改变.结果 正常组胆碱、甘油磷酸胆碱、β-葡萄糖、α-葡萄糖、甘油三酯相对浓度高于经DEN处理的肝脏肿瘤组(P〈0.05);DEN处理组的谷氨酸、异亮氨酸、赖氨酸的相对浓度高于正常对照组(P〈0.05),以上均具有统计学意义.结论 离体高分辨魔角旋转磁共振波谱分析方法及OPLS统计法可以敏感地检测出大鼠肝脏肿瘤的代谢改变.
Objective To study the metabolism of liver cancer model(Wistar rat liver tissues) and normal liver tissue in vitro at the molecular level. Methods 65 rats with 50 diethylnitrosamine (DEN)-induced liver cancer modes and 15 normal rats as control group underwent high resolution magic--angle spinning 1H MRS. The liver metabolism of the tumors and normal tissue in both ex- perimental group and control group was analysed with specialized software and orthogonal signal correction partial least squares (OPLS) method. The results were compared with that of pathology. Results The relative concentration of the choline,Glycerophos- phocholine, β- glucose, α- glucose, triglyceride in control group were higher than that in experimental group ( P〈 0.05 ). While the relative concentration of glutamate,isoleucine, lysine in experimental group were higher than that in control group( P〈0.05 ). Conclusion In vitro high resolution magic-angle spinning method of spectrum analysis and OPLS can sensitively detect metabolic changes in rat liver tumors.
