目的:对自发性高血压大鼠(spontaneously hypertensive rats,SHR)实施不同强度的运动干预,探讨中、高强度的运动对高血压大鼠心肌纤维化的影响及其可能的基质金属蛋白酶的表达基质。方法:选用3月龄雄性自发性高血压大鼠(SHR)24只,随机分为安静对照组(SHR-C)、SHR中等强度组(SHR-MEX)、SHR高强度组(SHR-HEX),正常雄性WKY大鼠(WKY-C)8只。SHR大鼠运动训练12周,60 min/day,5 d/周。运动强度:SHR-MEX组坡度0°,18~20 m/min;SHR-HEX组:坡度0°,25~28 m/min。隔周测量各组血压,12周后,测量全心质量指数、心肌蛋白胶原的表达以及心肌组织基质金属蛋白酶9(MMP-9)和基质金属蛋白酶组织抑制剂1(TIMP-1)的表达。结果:1)SHRMEX组的收缩压显著低于SHR-C组,SHR-HEX组的收缩压要显著高于SHR-MEX组;2)SHR-C组的全心质量指数显著高于WKY-C组,SHR-MEX组显著低于SHR-C组,SHR-HEX组显著高于SHR-C组;3)SHR组相比于SHR-C组心肌胶原容积分数、I、Ⅲ型胶原蛋白表达显著升高,SHR-MEX组相比于SHR-C组显著降低,SHR-HEX组相比于SHR-C组显著升高;4)SHR-C组相比于WKY-C组MMP-9以及MMP-1/TIMP-1显著升高,SHR-MEX组相比SHR-C组显著降低,SHR-HEX组相比SHR-C组显著升高。结论:中等强度运动对大鼠心肌纤维化的改善要好于高强度运动,其中MMP-9与TIMP-1的表达平衡可能是其重要机制之一。
Objective:Taking the medium and high intensity treadmill exercises to be an intervention on myocardial fibrosis in spontaneously hypertensive rats(SHR),the authors want to investigate the effect of this kind of intervention and to explore the underlying mechanism of expression of matrix metalloproteinase.Methods:24 males SHR rats were randomly divided into 3 groups:sedentary group(SHR-C),middle intense exercise group(SHR-MEX),high intense exercise group(SHR-HEX) and there were 22 males WKY rats(WKY-C).SHR-MEX underwent treadmill training at 18-20m/min for 60 minutes,5d/week,for 12 weeks.SHR-HEX underwent treadmill 25-28m/minutes,5d/week,for 12 weeks.After 12 weeks,blood pressure,heart mass index,the expression of myocardial collagen fiber,and the protein expression of matrix metalloproteinase-9 and tissue inhibitor of metalloproteinase-l were examined.Result:(1) The SBP in SHR-C group was significantly lower than that in SHR-C group,but the condition was quite opposite in SHR-HEX group whose SEP was significantly higher than the SEP in SHR-C group;(2) The heart mass index in SHR-C group was significantly higher than that in WKY-C group,the heart mass index in SHR-MED group was significantly lower than that in SHR-C group,but the heart mass index in SHR-HEX was higher than that in SHR-C group significantly;(3) The CVF,expression of Ⅰ,Ⅲ collagen fibril in SHR-C group was significantly higher than that in WKY-C group,and these index in SHR-MEX is lower significantly,compared with that in SHR-C group,the CVF,expression of Ⅰ、Ⅲ collagen fibril in SHR-HEX group was higher than that in SHR-C group significantly;(4) The MMP-9 and the rate between MMP-9 and TIMP-1 in SHR-C group were significantly higher than that in WKY-C group.Also,these indexes in SHR-MEX group were lower that these in SHR-C group.Oppositely,the MMP-9 and rate between MMP-9 and TIM P-1 in SHR-HEX group were higher than these in SHR-C group.Conclusion:the middle intensity of aerobic exercise has a b
