哺乳动物皮肤具有多种功能,这些功能是由基因决定的,而基因的表达要受到转录因子的调节。本研究通过参照人皮肤基因的转录因子分析羊驼皮肤的EST库,发现了8个转录因子和6个转录因子结合位点,它们调控着羊驼生活适应性、皮肤生理特征以及皮肤衍生物的生理特性等,其中重要的有调控羊驼黑色素细胞内决定其毛色的转录因子mierophthalmia-associated transcription factor(MITF)及其结合位点cyclic adenosine monophosphate response element-binding1(CREB-1),用实时荧光定量PCR法检测发现二者在不同羊驼毛色皮肤中的表达在转录水平存在显著差异,它们的生物学特点对研究羊驼皮肤中决定羊驼皮肤生理功能和羊驼毛色的分子机理提供了重要的理论基础。
Many functions of mammalian skin are decided by genes, which are expressed by the regula- tion of transcription factors. In order to find the mechanism of transcription factor expression, the EST library of alpaca skin was analyzed on the basis of the identification of transcription factor expression in hu- man skin. The results showed that 8 transcription factors and 6 transcription factor binding sites were found, which regulated the environmental adaptability, physiological characteristics and the derivatives's traits of alpaca skin. Especially, the hair color gene microphthalmia-associated transcription factor (MITF) and one of it's transcription factor cyclic adenosine monophosphate response element-binding (CREB-1) were expressed. Moreover, there was the significant difference of transcription expression in alpaca skin with different coat color by real-time PCR test. The data will help to study the molecular mechanism of the physiological characteristics and hair color of alpaca.
