目的:探讨辛芷鼻敏胶囊对变应性鼻炎(Allergic rhinitis,AR)大鼠的血清中白介素13(Interleukin-13,IL-13)和鼻粘膜组织的信号转导和转录激活因子-6(Signal transducers and activator s of transcription-6,STAT6) mRNA及蛋白表达,明确药效机制.方法:40只大鼠随机分为空白对照组、模型组、对照组、实验组,用卵清蛋白建立 AR 大鼠模型,通过对造模的大鼠灌服辛芷鼻敏胶囊及与同类药物对照,用酶联免疫吸附技术(Enzyme labeled immunosorbent assay,ELISA)定量测定各组大鼠血清IL-13水平,用原位杂交(ISH)、免疫组化方法半定量检测鼻粘膜中STAT6mRNA、STAT6蛋白含量的表达.结果:实验组大鼠血清的IL-13表达水平和鼻粘膜中STAT6mRNA、STAT6蛋白含量的表达比例明显低于模型组及对照组(P〈0.05).结论:辛芷鼻敏胶囊可能降低STAT6mRNA、STAT6蛋白含量表达,抑制大鼠AR发病过程中IL-13的表达水平,进而减少嗜酸性粒细胞浸润而控制症状.
Objective: To explore the effect of Xinzhi Bimin capsule on the content of IL-13 in blood serum and the expression of STAT6 mRNA and protein in nasal mucosa of rat models with allergic rhinitis and identify the mechanism of pharmacodynamic action. Meth- ods: Forty rats were divided into four groups at random, normal control group, model group, control group and experimental group. Ovalbuminsensitized rats used as animal of AR were taken orally with Xinzhi Bimin capsule and compared with the different model rat established in AR and the normal control group, the expression of positive cell of STAT6 mRNA and protein in nasal mucosa was detected in semi quantity by in situ hybridization(ISH)and immunohistochemical method and the amount of IL-13 in blood serum was detected in quantity by ELISA. Results: The proportion of positive cells to STAT6 mRNA and protein content in nasal mucosa and the level of IL-13 in blood serum were markedly lower in the experimental group than in the model group (P 〈 0.05) and the positive control group( P 〈 0.05) except the normal control group. Conclusion: The result indicates that the capsule of Xinzhi Bimin can reduce the expression of STAT6 mRNA and protein content in nasal mucosa of rats and down-regulate the content of IL-13 in serum and reduce the infiltration of EOS and control the symptoms of AR in rats.
