目的:观察中药复方草果知母汤对癫痫大鼠海马神经元超微结构的影响。方法:实验于2005-03/05在广西中医学院中心实验室和广西医科大学电镜室完成。选择雄性SD大鼠32只,随机数字表法分为4组,即正常组、模型组、中药组和西药组;除正常组外,其他组动物均以戊四唑慢性诱导复制癫痫模型,造模同时给药,中药组每天给予草果知母汤(草果10g,知母10g,厚朴10g,半夏10g,黄芩10g,甘草10g等,广西中医学院第一附属医院研制,批号:20050108)灌胃,5g/kg;西药组每天给予苯巴比妥混悬液灌胃,60mg/kg;模型组和正常组每天均以双蒸水灌胃2mL/kg;1次/d,持续5周。从第2周开始,每周末造模后1h,各组抽取2只大鼠,采用透射电镜观察海马区神经元超微结构。结果:纳入大鼠32只,实验过程中,模型组有2只大鼠因惊厥发作而死亡;中药组、西药组各有1只大鼠不明原因死亡,进入结果分析28只。①模型组海马区神经元超微结构损伤显著,从第3周开始,神经元数目逐渐减少,至第5周,海马区出现较多的暗细胞,胶质细胞数目增多,神经元出现髓样变性,周围的线粒体肿胀,线粒体棘及其上的颗粒明显减少,滑面内质网空泡化,粗面内质网膨胀,其上的核糖体数量减少,高尔基体肿胀,胞内溶酶体数量增多,染色质浓染,核仁浓缩、变小,突触的数量明显减少,突触间隙及前后突触体内的囊泡的数量明显减少。(参中药组、西药组神经元数目略减少,形态结构未见明显改变。(固正常组未见明显改变。结论:戊四唑慢性诱导癫痫发作可致大鼠海马区神经元超微结构明显损伤,草果知母汤具有良好地抗损伤作用。
AIM: To investigate the effect of tsaoko-anemarrhenae decoction, a Chinese medicine compound,on the uhrastructure of hippocampal neurons in epileptic rats. METHODS: The experiment was conducted in the central laboratory of Guangxi College of Traditional Chinese Medicine and Electron Microscope Room of Guangxi Medical University from March to May 2005. Totally 32 male SD rats were selected and randomly divided into 4 groups: normal group, model group, Chinese medicine group and western medicine group. Except rats in the normal group, all the rats were given pentylenetetrazol (PTZ) to induce models of chronic epilepsy, besides given the medicine. The rats in the Chinese medicine group were intragastrically infused with 5 g/kg tsaoko-anemarrhenae decoction (10 g tsaoko, 10 g anemarrhenae, 10 g magnolia bark, 10 g pinelliae, 10 g baical skullcap root and 10 g licorice root, manufactured by the First Affiliated Hospital of Guangxi College of Traditional Chinese Medicine, batch number: 20050108); the rats in the western medicine group were given 60 mg/kg Phenobarbital suspension; the model and normal groups were given 2 mL/kg double distilled water, all the medicine was given once daily for 5 weeks. Since the second week, after building a mold 1 hour at each weekend, 2 rats were santpled randomly from each group to observe the uhrastrncture of hippocampal neurons with transmission electron microscope. RESULTS: Of the 32 rats, except 2 rats of the model group died of eclampsia, 1 rat in the Chinese medicine group and 1 in the western medicine group died with unknown reasons, 28 rats entered the result analysis. ①Ultrastrncture of hippocampal neurons of the model group was damaged obviously.
