目的:观察中药复方草果知母汤对癫痫大鼠行为和脑海马神经元显微结构的影响。 方法:实验于2005-03/05在广西中医学院中心实验室完成。选择SD雄性大鼠110只,按随机数字表法分成4组,草果知母汤组、苯巴比妥组、模型组各34只(每组10只用于电镜),正常对照组8只(2只用于电镜)。除正常对照组,其他3组均以戊四唑慢性点燃癫痫模型。各组在造模同时即开始给药,草果知母汤组予草果知母汤液(由草果,知母,厚朴,半夏,黄芩,甘草等组成)5g/(kg·d)灌胃,苯巴比妥组予苯巴比妥混悬液60mg/(kg·d)灌胃,模型组和正常对照组均以双蒸水灌胃2mL/(ks·d);1次/d,持续5周。采用Smialowki 6级评分法进行行为学观察(1级,节律性点头或头部颤搐;6级,强直性惊厥),采用苏木精-伊红染色和光镜技术观察大鼠脑海马神经元的显微结构。 结果:110只大鼠全部进入结果分析,无脱失。①大鼠行为学观察结果:除正常对照组外,各组大鼠均出现惊厥行为,点燃率为83%。随着点燃周数的增加,惊厥级别不断增高,尤以模型组最为显著,第5周时草果知母汤组、苯巴比妥组惊厥级别均显著低于模型组[2.30±0.54,1.85±0.46,5.2±0.80(P〈0.05)]。②大鼠脑海马神经元数目及显微结构:从第4周开始,与正常对照组比较,模型组脑海马区神经元数目明显减少(P〈0.05-0.01);与模型组比较,草果知母汤组、苯巴比妥组脑海马区神经元数目显著增多(P〈0.05-0.01),后两组差异未见显著性意义(P〉0.05)。与正常对照组比较,模型对照组的神经元出现不同程度的变性、坏死,重者伴有神经元凋亡;草果知母汤组、苯巴比妥组改变不明显。 结论:草果知母汤对癫痫大鼠有明显的抗惊厥作用,其抗痫疗效与苯巴比妥相似,且能较好
AIM: To investigate the effect of a Chinese compound prescription, decoction, which is on the behavior and the microstructure of hippocampal neurons in epileptic rats. METHODS: The experiment was performed at the Central Laboratory, GuangXi Traditional Chinese Medical University from March to May 2005. A total of 110 SD male rats were selected and randomly divided into 4 groups: tsaoko-anemarrhenae decoction group, phenobarbital group and model group with 34 rats in each (10 rats in each group determined under electron microscope), normal control group with 8 rats (2 rats determined under electron microscope). Except normal control group, the pentylenetetrazol(PTZ)chronically induced epileptic SD rats were selected as the animal model, At the same time, the rats of each group were administrated. The 5g/kg tsaoko-anemarrbenae decoction (composed of tsaoko, anemarrhenae, magnolia bark, pinelliae tuber, baical skullcap root, radix and so on) was given to the rats in the tsaoko-anemarrhenae decoction group by gastric perfusion daily. The rats in the phenobarbital group were treated with 60 mg/kg phenobarbital suspension by gastric perfusion. Those in the model group and normal control group were treated with gastric, perfusion with 2 mL/kg double distilled water, once a day for 5 weeks. Behavioral observation was conducted with Smialowki 6-grade assessment (1 grade represented rhythmicity nutation or twitching in head; 6 grade represented tonic convulsion). The microstructure of hippocampal neurons of rats was observed with hematoxylin-eosin staining and light microscope technique. RESULTS: Totally 110 rats were involved in the result analysis, no dropout. ①The observational result of praxiology: Apart of the normal control group, all the groups appeared.epileptic behavior with the induced rate of 83%.
