目的观察nesfatin-1对脑干孤束核葡萄糖敏感神经元电活动影响,探讨其抑制摄食作用的可能机制。方法采用全细胞膜片钳技术,在电流钳下,记录nesfatin-1对孤束核葡萄糖敏感神经元电活动的影响。结果在孤束核中共记录到30个神经元,给予葡萄糖(5mmol/L)灌流后,有18个(60.0%)对葡萄糖有反应,其中13个放电频率升高(G-EXC,t=2.509,P〈0.05),5个放电频率下降(G-INH,t=12.79,P〈0.01),其余12个无反应。在13个G-EXC神经元,灌流nesfatin-1(10nmol/L)后12个放电频率增加(t=3.346,P〈0.01),1个无反应。在5个G-INH神经元,灌流nesfatin-1(10nmol/L)后4个放电频率减少(t=11.877,P〈0.01),1个无反应。结论 nesfatin-1能够调制孤束核葡萄糖敏感神经元的兴奋性,这可能是nesfatin-1作用于孤束核抑制摄食的机制之一。
Objective To observe the effects of nesfatin-1 on glucose-sensitive neurons in nucleus tractus solitarius(NTS),and explore the possible mechanisms of NTS in controlling food intake.Methods Whole-cell patch-clamp technique was used to record the discharges of NTS induced by nesfatin-1 administration.Results Thirty NTS neurons were treated with glucose,of which 18(60.0%)responded to glucose with 13 as glucose-excited(G-EXC)neurons and five as glucose-inhibited(G-INH)neurons.The other 12 showed no response.Nesfatin-1 administration(10 nmol/L)increased the firing rates of 12 G-EXC neurons(12/13)(t=3.346,P〈0.01),but decreased the firing rates of four G-INH neurons(4/5)(t=11.877,P〈0.01).One was found with no response.Conclusion Nesfatin-1 can regulate the activity of G-EXC neurons,which might be one of the mechanisms of NTS controlling food intake.
