目的研究大骨节病可疑致病因子NIV毒素和保护因子硒对体外培养软骨细胞透明质酸代谢的影响,为阐明NIV毒素损伤软骨细胞的机制提供实验依据。方法本实验以体外培养的人胚软骨细胞、培养液为研究对象,利用RT—PCR方法检测透明质酸合成酶-2mRNA在软骨细胞中的转录情况;利用放射免疫沉淀技术检测细胞培养液中透明质酸含量。结果NIV毒素作用下软骨细胞中透明质酸合成争2mRNA水平升高,毒素加硒组与毒素组基因表达水平无显著性变化。NIV毒素组培养液中透明质酸的含量显著增加。毒素加硒组与毒素组变化趋势一致,但数值略下降。结论NIV毒素对于软骨细胞的透明质酸代谢具有明显的干扰作用,进而引起软骨细胞外基质蛋白聚糖等组分代谢紊乱;补硒能够拮抗NIV对软骨细胞的损伤,但作用有限。
Objective:The purpose of the study is m discover the effect of nivalenol and selenium on the metabolism of hyaluronic acid, in order to explore the role of the suspected etiological factor in the process of Kashin- Beck Disease. Method Hyaluronan synthase- 2 mRNA expression was measured by RT- PCR and radioimmunoprecipitation method was used to detect the levels of hyaluronic acid in culture medium. Result NIV increased hyaluronan synthase- 2 mRNA expression. After Se was added, the mRNA levels did not change significantly compared with the groups without Se. The content of hyaluronic acid in medium with NIV was higher than that in other groups. Conclusiva NIV has a significant effect on chondrocytes cultured in vitro, it can perturb hyaluronic acid synthsis and metabolism, which eventually lead to the metabolic disorder of the cartilage matrix components. Supplying Se can partially alleviate the damage of NIV on cartilage.
