目的:探讨切割穹隆海马伞海马中具有诱导神经干细胞向神经元分化生物活性的83ku差异蛋白的组成成分及其功能。方法:切割sD大鼠穹隆海马伞后14d海马进行非变性聚丙烯酰胺凝胶电泳(Native PAGE),应用电喷雾质谱分析、蛋白质数据库和文献分析等方法对83ku差异蛋白进行检测和功能分析。结果:质谱分析找到了17组肽段,这些蛋白按照其功能可以分为:细胞骨架蛋白、参与代谢的酶、信号传递、蛋白降解、氧化应激、神经递质运输、突触形成、功能未知蛋白。结论:切割穹隆海马伞海马83ku差异蛋白可以通过参与Rho信号通路,调控神经干细胞向神经元分化。
Objective: To explore the ingredients and functions of the active 83 ku proteins inducing NSCs to differentiate into neurons from the fimbria/fornix transected hippocampus. Methods: The Sprague- Dawley rats' hippocampi on the 14th day after fimbria/fornix transection were used for Native-PAGE. Electrospray ionization mass spectrometry, protein data bank and bibliometrics were applied to identify the 83 ku proteins and analyze their functions. Results: Mass spectrometry analysis of 83 ku protein found 17 groups of peptides which were divided into several functional groups: cytoskeletal protein, enzyme in metabolism, signal transduction, protein degradation, oxidative stress, neurotransmitter transport, synapse formation and unknown function protein etc. Conclusion: 83 ku proteins in the fimbria/fornix transected hippocampus may regulate the differentiation of NSCs into neurons via Rho pathway.
