目的:观察异基因外周血干细胞移植供者动员前后淋巴细胞亚群及造血干细胞的变化。方法:粒细胞集落刺激因子(rhG-CSF)动员外周血,直接免疫荧光法标记淋巴细胞和CD34细胞,用流式细胞仪检测其表达情况,甲基纤维素半固体培养基进行CFU-GM培养评价干细胞克隆生成能力。结果:rhG-CSF动员后淋巴细胞亚群与动员前相比无明显变化,CD34+细胞由动员前的0.12±0.08增加到0.83±0.26,CFU-GM有动员前的12±6增加到102±16。结论:正常供者外周血动员后CD34+细胞明显增加,淋巴细胞无明显变化。
Objective :To investigate the alteration of donor's lymphocyte subpopulation and haemopoietic stem cell before and after rhG-CSF mobilization in allogenic peripheral blood stem cell transplantation. Methods: Before and after recombinant human Granulocyte-Colony Stimulating Factor (rhG-CSF) mobilization, lymphocytes and CD34 ~ cells were stained directly and counted by flow cytometry. Stem cell clone proliferation was estimated by CFU-GM with methylcellulose semisolid medium. Results:Before rhG-CSF mobilization, the percentage of CD34 ^+ cells was 0.12 ±0.08 and CFU-GM was 12 ±6. After rhG-CSF mobilization the percentage of CD34 ^+ cells increased to 0.83 ± 0.26 and CFU-GM increased to 102 ± 16. Tymphocyte subpopulation didnt change significantly after rhG-CSF mobilization. Conclusion:Mter rhG-CSF mobilization, the percentage of donor's CD34 ^+ cells increases greatly and lymphocyte subpopulation doesn't change significantly.
