本文通过对传统的化学合成方法进行改进,在反应液中加入保险粉,且以柠檬酸-磷酸氢二钠缓冲溶液替代水溶液,以高收率合成得到较高纯度的5-甲基四氢叶酸。利用高效液相色谱检测手段监测反应,确定硼氢化钠的添加量、反应温度、缓冲液浓度和p H等因素对反应的影响。结果表明,叶酸为0.2g、硼氢化钠的总添加量为0.65g、甲醛添加量为0.4m L、缓冲液浓度为0.2mol/L、p H为6.5和反应温度为50℃时,目标产物收率达70.4%,纯度为88.9%。
In this paper,we have established a novel method to synthesize 5-methyltetrahydrofolate( 5-MTHF)with high purity and high yield by introducing sodium hyposulfite into the reaction and also substituting citric aciddisodium hydrogen phosphate buffer solution for water. HPLC was used to investigate the influential factors of the reaction,including the addition of sodium salt,reaction temperature,buffer concentration,p H values,etc. The results revealed that,when the folic acid amount was 0. 2g,Na BH4 amount was 0. 65 g,the reaction temperature was50℃,the concentration of the buffer was 0. 2mol / L,p H value was 6. 5 and the addition of formaldehyde was 0. 4m L,over 88. 9% purity of the target product was obtained with the yield of 70. 4%.
