目的建立一测多评(QAMS)的泽泻多指标质量控制方法,并验证该方法在泽泻含量测定中应用的可行性和技术适应性。方法采用HPLC,以25-乙酰泽泻醇B为内参物,计算泽泻醇A、24-乙酰泽泻醇A和泽泻醇B的相对校正因子,并利用该校正因子计算含量,同时与采用外标法测定4个三萜类成分的含量进行比较。结果建立的校正因子重现性良好,31批泽泻药材中4个三萜类的含量计算值与外标法实测值间(RSD≤3.52%)无显著差异。结论本实验所建立的一测多评法稳定、准确,可用于泽泻三萜类成分的质量控制。
OBJECTIVE To develop a simultaneous quantitative analysis method of four triterpenoids in Alismatis Rhizoma by multi-components by single maker (QAMS). METHODS Four main triterpenoids ( alisol A, alisol A 24-acetate, alisol B and alisol B 23-acetate) were selected as analytes to evaluate the quality of Alismatis Rhizoma. Alisol B 23-acetate was used as the internal refer- ence standard and three relative correction factors (RCFs) to alisol B 23-acetate were calculated. These RCFs were obtained by high performance liquid chromatography coupled with diode array detector under various chromatographic conditions. Then 31 batches of A1- ismatis Rhizoma samples collected from different areas were determined by both external standard method (ESM) and QAMS. RE- SULTS The RCFs were obtained with good reproducibility ( RSD ≤ 2.25% ). No obvious differences were observed between the analysis results of QAMS method and ESM method ( RSD ≤3.52% ). CONCLUSION The established QAMS method is reliable and feasible for simultaneous analysis of four triterpenoids and could be used for quality control of Alismatis Rhizoma.
