目的:研究细胞外基质(ECM)对哮喘大鼠气道平滑肌细胞(ASMCs)磷脂酰肌醇-3-激酶p85(P13Kp85)表达的影响。方法:卵白蛋白(OVA)致敏法复制大鼠哮喘模型,将培养后的大鼠叶支气管ASMCs分别种植在涂覆有纤维连接蛋白、I型胶原及层粘蛋白的培养瓶中培养,Western blot检测各组ASMCs中P13Kp85蛋白表达,Realtime PCR检测其mRNA。另取12只正常大鼠做对照。结果:哮喘纤维连接蛋白组、层粘蛋白组、Ⅰ型胶原蛋白组ASMCs中P13Kp85 mRNA的表达分别是(7.63±0.61)、(9.73±0.68)、(16.22±0.88),均明显高于对照组(5.93±0.75)(P〈0.01);蛋白表达情况与mRNA趋势一致。结论:纤维连接蛋白、Ⅰ型胶原及层粘蛋白可以上调哮喘大鼠气道平滑肌细胞中P13Kp85表达。
Objective To investigate the influence of extracellular matrix(ECM) protein on the expression of phosphatidylinositol 3-kinase p85(PI3Kp85) of airway smooth muscle cells(ASMCs) in asthmatic rats.Methods The rats model of asthma were induced with OVA injection,the cultured ASMCs of rats lobe bronchus were implanted into flasks treated with fibronectin,collagen-Ⅰ and laminin.The expression of PI3Kp85 protein was determined with Western blotting,and the mRNA expression of PI3Kp85 was detected by real-time PCR.Another 12 rats were selected as healthy controls.Results The expression of PI3Kp85 mRNA in asthmatic group treated with fibronectin,collagen-Ⅰ and laminin were 7.63±0.61,9.93±0.12,16.22±0.88,respectively.They were all significantly higer than that of control group(5.93±0.75)(P0.01).The expression of PI3Kp85 protein was similar to PI3Kp85 mRNA.Conclusion Fibronectin,collagen-Ⅰ and laminin could up-regulate the expression of PI3Kp85 in ASMCs of asthmatic rats.
