目的:探讨益气化瘀方对膝骨性关节炎模型小鼠异位软骨内成骨的影响。方法:12周龄C57BL/6雄性小鼠30只,随机分为假手术组、模型组和益气化瘀方组,每组10只。模型组和益气化瘀方组小鼠通过左膝关节行内侧半月板及前交叉韧带离断术诱导膝骨关节炎,并分别给予0.9%氯化钠溶液和益气化瘀方(0.1m L/10g)灌胃。8周后收集小鼠血清和左侧膝关节。组织学染色观察左侧膝关节异位软骨内成骨情况。免疫组织化学染色观察ColⅡ、Col X、RUNX2、IL-1β的表达情况,ELISA法检测血清IL-1β水平。结果:与模型组比较,益气化瘀方组膝关节半月板钙化和骨赘形成明显减轻,半月板中ColⅡ表达增加,Col X、RUNX2表达降低,小鼠血清中和异位骨化处IL-1β表达均下调。结论:益气化瘀方可以抑制膝骨性关节炎模型小鼠异位软骨内成骨,可能与抑制IL-1β表达有关。
Objective: To explore the inhibitory effect of Yiqi Huayu Formula on ectopic endochondral ossification in mice model of knee osteoarthritis. Methods: Twelve weeks of C57BL/6 male mice were randomly divided into the sham group, model group and Yiqi Huayu Formula group. Mice in model group and Yiqi Huayu Formula group were performed with anterior cruciate ligament transection and partial medial meniscectomy in the left knees to induce knee osteoarthritis, and were respectively gavaged with saline and Yiqi Huayu Formula. Eight weeks later, serum and the left knee joints were collected. Histological staining was performed to observe the ectopic endochondral bone formation of the left knee joints. Immunohistochemistry staining was used to detect proteins expression of Col II, Col X, RUNX2 and IL-1β. ELISA assay was used to detect the level of IL-1β in serum. Results: Compared with the model group, meniscus calcification and osteophyte formation in knee joints were relieved in Yiqi Huayu Formula group. Meanwhile, compared with the model group, the expression of Col II in meniscus was increased, while Col X and RUNX2 were decreased as well as the level of IL-1βin serum and heterotopic ossification region. Conclusion: Yiqi Huayu Formula could inhibit ectopic endochondral ossification in mice model of knee osteoarthritis, which may be related to an inhibition of IL-1β.
