目的 建立HPLC定量分析防己黄芪汤(防己、黄芪、白术和甘草)中防己诺林碱、粉防己碱、毛蕊异黄酮葡萄糖苷、甘草苷和甘草酸5种成分的方法.方法 采用HPLC外标法,样品经过50%甲醇提取,采用DIKMA DIAMONSIL (2)C18柱,甲醇-0.2 mol/L乙酸铵0.1%甲酸水溶液为流动相,梯度洗脱,紫外检测波长254 nm和270 nm.结果 防己诺林碱、粉防己碱、毛蕊异黄酮葡萄糖苷、甘草苷和甘草酸分别在4 ~ 50 μg/mL、3~ 222 μg/mL、7~111 μg/mL、15 ~ 244 μg/mL、14 ~ 222μg/mL范围内呈良好线性关系.结论 本方法准确,可同时测定防己黄芪汤的3种药材中的5种成分.
AIM To establish a quantitative analysis method in determining five constituents (fangchinoline,tetrandrine,calycosin-7-glucoside,liquiritin,and glyeyrrhizic acid) in Fangji Huangqi Decoction (Stephaniae tetrandrae Radix,Astragali Radix,Atractylodes Macrocephalae Rhizoma,and Glycyrrhizae Radix et Rhizoma).METHODS The separation of 50% ethanolic extract of Fangji Huangqi Decoction was performed on a DIKMA DIAMONSIL (2) C18 column.The mobile phase consisted of methanol-0.2 mol/L ammonium acetate,0.1% formic acid for a gradient elution,and the detection wavelength was set at 254 nm and 270 nm,respectively.Each content of five constituents was calculated by external standard method.RESULTS The linear ranges of fangchinoline,tetrandrine,calycosin-7-glucoside,liquiritin,and glyeyrrhizic acid were 4-50 μg/mL,3-222 μg/mL,7-111 μg/mL,15-244 μg/mL,14-222 μg/mL,respectively.CONCLUSION HPLC method is suitable for simultaneous determination of five constituents accurately from three herbs in Fangji Huangqi Decoction.
