目的:评价核基质结合区结合蛋白与恶性黑素瘤的相关性。方法:以取材自两个健康供者的原代培养黑素细胞和乳癌细胞系MDA—MB一231作为对照,应用RT—PCR方法,检测部分与肿瘤相关的核基质结合区结合蛋白(SMARl、LaminB1、Nucleohn、hnRNPU、SATBl、YBXl、CUXl和LaminB2)mRNA在恶性黑素瘤细胞系A375、M14和MV3的表达。结果:所有被检测核基质结合区结合蛋白在正常黑素细胞均未见表达;在三个恶性黑素瘤细胞系均有表达的有SMARl、LaminB1、Nucleolin、hnnNPU、SATBl和YBXl,以Nucleolin、hnRNPU和SATBl的表达水平与正常黑素细胞的差异较大。乳癌细胞系MDA—MB一231与黑素瘤细胞表达谱相似。结论:Nucleolin、hnRNPU和SATBl等MARBP可能参与了黑素瘤的发生与发展。
Objective: To investigate the expression of matrix attachment regions binding proteins (MARBPs) in maliganant melanoma. Method: The expression of ten MARBPs (SMAR1, CUX1, Lamin B1, Nucleolin, hnRNP U, SATB1, YBX1 andLamin B2) in three melanoma cell lines (A375, M14 and MV3) was measured by RT- PCR, with normal primary cultured melanocytes from two healthy donors and breast cancer cell line MDA- MB - 231 as controls. Results: SMAR1, Lamin B1, Nucleolin, hnRNP U, SATB1, and YBX1 were over- expressed in these three melanoma cell lines, especially Nucleolin, hnRNP U, SATB1, similar to MDA - MB - 231, whereas no MARBP was detected in normal melanocytes. Conclusion: These results suggest that some MARBPs, such as Nu- cleolin, hnRNP U and SATB1, may play roles in the development of malignant melanoma.
