目的建立吗啡诱导的斑马鱼条件性位置偏爱模型,探讨诱导斑马鱼形成位置偏爱的最佳条件。方法根据条件性位置偏爱(CPP)原理和斑马鱼的生物学特性,对斑马鱼的天然偏爱进行测定后,腹腔注射不同浓度的吗啡,并与非偏爱箱(伴药箱)进行搭配;随之,斑马鱼注射等体积生理盐水,与偏爱箱进行搭配。经过连续3次的训练,观察吗啡诱导的斑马鱼CPP效应。 结果斑马鱼注射40-50 μg·g^-1吗啡并进行连续3次CPP训练,能诱导其在伴药箱中的活动时间增加,与训练前相比,差异有显著性(P〈0.01),与生理盐水组相比,差异也具有显著性(P〈0.01)。结论斑马鱼进行腹腔注射给药40-50 μg·g^-1吗啡,并进行连续3次训练,能形成明显的CPP。
Aim To establish the morphine-induced conditioned place preference ( CPP ) model and explore the optimum condition of the CPP formation in zebrafish. Methods According to the principle of CPP and the biological characteristics of the zebrafish,after determination of the natural preference of the zebrafish,the zebrafish was given morphine of different concentrations by intraperitoneal injection, and then it was matched with non-preference compartment; subsequently, the zebrafish was given equal volume of saline and then matched with preference compartment. Morphine-induced CPP effect on zebrafish after three consecutive trainings were observed. Results After being given 40 -50 μg · g^-1 morphine by intraperitoneal injection and three consecutive trainings, we found that the activity time of zebrafish in non-preference compartment was increased significantly. Compared with pre-training or saline group, there was a sig- nificant difference. Conclusion The zebrafish shows significant place preference by intraperitoneal injection of 40-50 μg · g^-1 morphine and three consecutive trainings.
