目的观察补肾通脉方对伴胰岛素抵抗(insulin resistance,IR)的多囊卵巢综合征(polycystic ovarian syndrome,PCOS)大鼠肝脏和脂肪组织中磷脂酰肌醇-3激酶(PI-3K)p85a亚基蛋白表达的影响。方法23日龄雌性SD大鼠皮下注射硫酸普拉睾酮钠9mg/(100g·d),共20d,联合高脂饮食喂养80d建立IR的PCOS大鼠模型,成模大鼠随机分为模型组和治疗组,另设15只为正常对照组。治疗组大鼠以补肾通脉方进行干预。光镜下观察大鼠排卵情况;通过葡萄糖氧化酶法测定空腹血糖(FPG);以放射免疫法测定空腹血清胰岛素(FINS);采用Western blot技术检测肝脏和脂肪组织中PI-3Kp85d的蛋白表达。结果与模型组比较,治疗组平均排卵数及排卵率明显增加(P〈0.01),FINS浓度显著降低(P%0.01)。治疗组肝脏和脂肪组织中PI-3Kp85a的蛋白表达水平均较模型组明显增加(P〈0.01.P〈0.05)。结论补肾通脉方可改善伴IR的PCOS大鼠的胰岛素抵抗,其机制可能与其上调肝脏和脂肪组织中PI-3K p85α蛋白的表达有关。
Abstract Objective To obverse the effects of Bushen Tongmai Recipe(BSTMR) on protein expression of phosphoinositide-3 kinase (PI-3K) p85α in hepatic and adipose tissues in PCOS rats accompanying with insulin resistance (IR). Methods 23 day-old female SD rats were injected subcutaneously with sodium prasterone sulfate 9 rag/(100 g · d) for 20 days,and fed with high-fat forage for 80 days to induce PCOS rats accompanying with IR. Then the rats were randomly divided into the model group and the treated group. Meanwhile, a group of fifteen rats of the same age was considered as the normal control group. The treated group were administered with BSTMR. The ovulation situations were examined by light microscope. Fasting plasma glucose (FPG) was determined using glucose oxidase method. Serum fasting insulin (FINS) was determined by radioimmunoassay (RIA). Western blot analysis was employed to detect protein expression of PI-3K p85u in hepatic and adipose tissues. Results Compared with the model group,the mean number of corpus luteum and the rate of ovulation in the treated group increased significantly (P〈0.01). The level of Fins in the treated group was greatly lower than that in the model group (P〈0.01). Protein expressions of PI-3K p85α in hepatic and adipose tissues were up-regulated significantly in the treated group (P〈0.01,P〈0.05). Conclusion BSTMR could improve insulin resistance in PCOS rats accompanying with IR,its molecular mechanisms might be closely related with the elevation of protein levels of PI-3K p85α in hepatic and adipose tissues.
