采用RNA-seq技术对志贺菌耐药性与非编码RNA之间的关系进行研究.首先采用梯度剂量的环丙沙星诱导福氏志贺菌2a标准菌株产生耐药性而获得耐药菌株,然后提取标准菌株与耐药菌株的总RNA进行RNA-seq分析.结果显示,在福氏志贺菌的2个样品中共发现100个候选sRNA,并与sRNA的数据库比对得到注释信息,每个候选sRNA都对应着很多个靶标基因,而且靶标基因的表达也有明显差异.预测这些sRNA可能会通过调控耐药基因和一些与细胞增殖或凋亡相关的基因发挥作用,因此还需要挑选合适的候选sRNA并对其用荧光定量PCR实验进行验证.
Relationship between drug resistance and non-coding RNA in Shigella was studied adopting RNA-seq technology,firstly standard Shigella flexneri 2a was induced with gradient dosage of ciprofloxacin to produce a drug-resistant S.flexneri,and then extracted total RNA of standard strain and drug resistant strain to conduct RNA-seq analyses.The results found 100 candidate sRNAs from two samples of Shigella,and got their annotation information when comparing with sRNA database,each candidate corresponds to a number of target genes,and the expression of target gene was significantly different.It was predicted that these sRNAs may play roles through regulation of drug resistance gene and some genes associated with cell proliferation or apoptosis,therefore,it still needs to choose the right candidate sRNA to test and verify them with fluorescence quantitative PCR.
