试验对烟碱降解能力强的节杆菌K15发酵条件的优化进行了研究,通过涂平板计数的方法得到节杆菌K15的最适发酵条件:淀粉、牛肉膏和硫酸镁分别作为节杆菌K15的最适碳源、氮源和无机盐,优化的培养基配方为6%的淀粉、2%的牛肉膏和0.2%的硫酸镁;最适pH值为7.5,最适温度为25℃,300mL瓶中加80mL的培养液的通气条件最佳,培养32h至48h为最适的菌种收获期。该研究结果为该菌株扩大培养以及工厂化生产提供了参考。
The ferment condition of Arthrobacter K15 which had the ability to degrade the nicotine was studied through smearing the culture medium statistical method. The result showed that starch, the beef ointment and MgSO4 were as the optimum carbon sources, nitrogen sources and inorganic salt of Arthrobacter K15, the optimum fermentation medium was 6% starch, 2% beef ointment and 0. 2% MgSO4 ; the optimum pH was 7.5, the temperature was 25 ℃, and 300 mL flask with 80 mL culture fluid was best aerated condition. The optimum harvest period of this kind of bacteria was 32 h to 48 h during the culture. The results provide reference for amplification culture of Arthrobacter K15 and in- dustrial production.
