高速逆流色谱法连续进样,分离制备杭白菊中的小白菊内酯。采用高速逆流色谱法进行分离,以正已烷-乙酸乙酯-甲醇-水(5∶5∶6∶4,V/V)为溶剂系统,上相为固定相,下相为流动相,连续进样3次,浸膏进样量330、350、370 mg,转速800 r/min,流速2.0 m L/min,检测波长254 nm。从杭白菊超声提取物中,分离得到小白菊内酯5.0、5.2、5.5 mg,纯度分别为93.1%、92.6%、92.9%。利用连续进样的高速逆流色谱法,可高效分离和纯化杭白菊中的小白菊内酯。
We separated and prepared parthenolide from Chrysanthemum morifofium with continuous multiple preparation and high-speed counter-current chromatography (HSCCC). We acquired parthenolide of 5.0,5.2 and 5. 5 mg from Chrysanthemum morifolium extract of 330,350 and 370 mg. Their purities were respectively 93.1%, 92.6%, and 92.9%. Experimental conditions were HSCCC, solvent system of n-hexane-ethyl acetate-methanol-water (5:5:6:4, V/V), upper stationary phase and lower mobile phase, rotation speed of 800. 0 r/min, flow rate of 2.0 mL/min and detection wavelength of 254 nm. It shows that continuous preparation HSCCC can efficiently separate and purify parthenolide from Chrysanthemum morifolium.
