以磺基水杨酸为显色剂,在p H为3的溶液中,建立了以分光光度法测定小鼠肝脏中铁(Ⅲ)含量的测定方法。在波长为500nm处测量体系吸光度,得到的铁(Ⅲ)标准曲线为A=6.894 3C(mg·L^-1)-0.1233,其相关系数为0.9997。实验所得小鼠肝脏铁(Ⅲ)含量与微波消解-火焰原子吸收光谱法测得的数据基本一致,均在测定结果范围之内,加标样品平均回收率为95.16%-103.73%。该方法测定快捷,简便,易操作,干扰少,需样品量少,所测络合物稳定性强。实验结果显示,相对于现存分析方法,此法可更加准确便捷地对小鼠肝脏铁含量进行测定。
The determination method of spectrophotometric determination of liver iron(Ⅲ) content was developed in sulfosalicylic acid as chromogenic reagent at p H 3. The absorbance of the system was measured at 500 nm,the Fe(Ⅲ) standard curve was A =6. 894 3C( mg·L^- 1)- 0. 123 3,the correlation coefficient of iron(Ⅲ) was 0. 999 7. The obtainedexperimentalliveriron( III) content wereconsistent withthe measured data of Microwave Digestion- Flame Atomic Absorption Spectrometry,the resultswerewithinthe range ofmeasurement,and the average recovery was 95. 16% - 103. 73%. This method is quick,simple,easy operation,less interference,small sample volume,measured by the complex stability. The experimental results show that,compared with the existing analysis method,this method can be more accurate to determine the content of iron in liver easily(Ⅲ).
