目的通过饮食控制碘铁摄入量,建立碘铁联合缺乏的大鼠动物模型。方法选择健康SPF/VAF级初断乳SD雄性大鼠22只,按体重随机分为对照组(饲以改良的AOAC饲料,碘含量为280μg/kg,铁含量为50mg/kg),缺碘缺铁组(饲以改良的AOAC缺碘缺铁饲料,碘含量为66μg/kg,铁含量为15mg/kg),每组11只。饲养4周后,测定体重和甲状腺重量,并计算甲状腺系数。测定大鼠尿碘、血清甲状腺激素、血红蛋白、血清铁水平和总铁结合力。结果缺碘缺铁组大鼠皮肤苍白,皮毛粗糙稀疏。与对照组比较,缺碘缺铁组大鼠体重、尿碘、血红蛋白、血清铁、血清游离三碘甲腺原氨酸(fT5)、游离甲状腺素(fT4)水平均较低,甲状腺重量及其脏器系数、总铁结合力和fT3/几比值均较高,差异有统计学意义(P〈0.05);促甲状腺激素(TSH)水平有升高趋势,但差异无统计学意义。结论该模型同时具有碘缺乏和铁缺乏特征,通过饮食控制大鼠碘铁摄入量成功制备碘铁联合缺乏大鼠动物模型。
Objective To establish a rat model with iodine and iron deficiency by controlling the dietary iodine and iron. Methods Twenty-two healthy SPF/VAF weaning male SD rats were randomly divided into control group(fed with modified AOAC feedstuff, in which iodine content was 280 μg/kg, and iron content was 50 mg/kg) and model group (fed with modified iodine and iron deficient AOAC feedstuff, in which iodine and iron content were 66 μg/kg and 15 mg/kg) by body weight,and 11 in each group. After 4 weeks of treatment, the body weight and thyroid glands weight were determined, and the index of thyroid gland was calculated. Urine iodine, thyroid hormone in the serum, hemoglobin concentration, serum iron and total iron binding capacity(TIBC) were determined. Results Rats in the model group showed pale skin, sparse rough fur. Compared with the control group, the body weight, the urine iodine, hemoglobin concentration, serum iron concentration and the free T3 and T4 of the rats in the model group were significantly lower (P〈0.05) and the weight and organ index of thyroid glands, TIBC and the ratio of T3/T4 was significantly higher (P〈0.05). TSH of the rats in the model group showed increasing trend. Conclusion This model has the characteristics of both iodine deficiency and iron deficiency at the same time, so it is a successful rat model with combined iodine and iron deficiency established by controlling dietary iodine and iron.
