透明质酸(hyaluronic acid,HA)是一种细胞外基质黏多糖,主要参与维持细胞外基质结构,调节细胞的多种生物学功能。近年来许多研究发现,肿瘤发生时人体内的HA平衡发生显著改变,不仅包括HA含量的变化,同时HA分子量大小也出现差异,从而导致一系列生物学功能的改变。因此,明确聚分子透明质酸(native high molecular weight hyaluronan,nHA)和寡分子透明质酸(oligosaccharides of hyaluronan,oHA)分别在肿瘤发生、发展、检测以及治疗中的作用及机制,对于肿瘤研究具有重要意义。本文主要针对nHA、oHA表达水平与肿瘤诊断的相关性,以及nHA和oHA在肿瘤发生、耐药以及靶向治疗应用中的研究进展作一综述。
Hyaluronan (HA) is a kind of mucopolysaccharide existing in extracellular matrix, playing a critial role in maintaining the extracellular matrix structure and regulating a variety of cellular functions. Recent studies found that the balance of HA changes when tumor occurs, not only the concentration of HA fluctuates, but also the molecular weight of HA alters, resulting in a series of changes of cellular functions. Therefore, it is of great significant to explicit the mechanism of native high molecular weight hyaluronan (nHA) and oligosaccharides of hyaluronan (oHA) invovled in tumor initiation, development, examination and treatment. This article is focused on the relationship between the expression of nHA/ oHA and the diagnosis of tumor. Another focal point of this article is advances in the role of HA in tumorigenesis, drug resistance and targeted therapy.
