研究旨在于运用体细胞克隆技术生产广西德保黑猪,并运用微卫星技术进行克隆猪胎儿的亲缘鉴定。试验选择广西德保黑猪体细胞作为核移植供体,巴马小型猪作为核移植受体猪;并选择9个微卫星位点以作克隆猪的亲缘鉴定。结果显示受体猪在夏季移植时受孕但流产,而冬季移植时均无受孕;微卫星分析也显示流产胎猪与供核成纤维细胞的亲缘关系一致。结果表明德保黑猪的体细胞克隆是可行的。
The objective of this study was to clone Guangxi Debao black pig using the SCNT (somatic cell nuclear transfer) technology and to identify the genetic relationship of clone pig fetus by microsatellite analysis. Somatic cells from Guangxi Debao black pig were used for the nuclear donor cells and Bama miniature pigs were used for the receptor pigs during production of cloned pigs. Nine microsatellite loci were used to identify the genetic relationship of clone pig. Our results showed that the receptor pig was successfully pregnant but aborted in summer. However, the receptor pigs got pregnancy failure after embryo transfer in winter. Microsatellite analysis results showed that genetic relationships were consistent between the aborted fetal pig and its nuclear donor cell. These results indicated that Guangxi Debao black pig was successfully cloned by SCNT technology.
