目的 观察姜黄素对培养的人血管内皮细胞内皮脂酶表达的影响,并探讨其可能的作用机制。方法 不同浓度的姜黄素处理HUVEC-12细胞。RT—PCR检测内皮酯酶(endothelial lipase,EL)mRNA的表达;Westernblot检测核因子-κB抑制因子-α(inhibitor of nuclear factor—κB-α,κB-α)蛋白的表达;间接免疫荧光检测核因子-κB(nuclear factor—κB,NF—κB)蛋白的活化。结果 IL-1β处理HUVEC-12细胞可以明显上调EL mRNA的表达,同时降低胞质蛋白IκB-α的表达水平,激活核转录因子NF-κB,增加胞核蛋白NF—κB的水平。姜黄素预处理HUVEC-12细胞可以抑制IL-1β对EL的上调作用,同时逆转IL-1β诱导的IκB-α蛋白降解和NF—κB活化。结论 姜黄素可以通过阻断NF—κB活化减少IL-1β诱导的人血管内皮细胞EL的表达。
Aim To investigate the effect and the possible mechanisms involved in the effect of curcumin on endothelial lipase (EL) expression in human umbilical vein endothelial cells line-12 (HUVEC-12). Methods HUVEC-12 were pretreated with different concentri-tions of curcumin (0- 100 ted with or without IL-1β. μmol·L^ -1 ) and then treated with or without IL-1β.eNODTHELIAL LIPASE(EL) expression was detected by RT-PCR analysis.The expression of inhibitor of nuclear factor-κB -α(IκB-α) was assayed by Western blot .Nuclear factor-κb(NF-κB) was analyzed by Immunofluorescence. Results IL-1β significantly induced EL expression at mRNA level as compared with control group. The IL-1β induction of EL expression was accompanied by activation of NF-κB and reduction of IκB-α. Cureumin almost completely blocked IL-1β-induced EL expression with a marked reduction of NF-κB and significant induction of IκB-α. Conclusion Curcumin inhibited IL-1β-induced EL expression via blocking NF-κB pathway in HUVEC-12 cells.
