目的探讨过氧化氢(H2O2)体外诱导细粒棘球蚴原头节细胞凋亡、Caspase-3表达和细胞超微结构的影响。方法RPMI1640添加谷氨酰胺组即体外培养细粒棘球蚴原头节,用5mmol/LH2O2诱导8h,使其发生细胞凋亡。用原位末端脱氧核糖核苷酸转移酶标记技术(TUNEL法)检测原头节细胞凋亡情况,用过氧化物酶标记链霉卵白素(SP)染色半胱天冬氨酸蛋白酶-3(caspase-3)。在透射电镜下观察原头节细胞超微结构的变化。结果过氧化氢诱导原头节细胞凋亡细胞增加,caspase-3表达增加,电镜观察原头节细胞异染色质增加,部分细胞染色质异常浓缩呈现凋亡细胞征象。结论Hz0z可诱导细粒棘球蚴原头节细胞凋亡,且caspase3参与原头节细胞的凋亡。
To investigate the effect of hydrogen peroxide (H2O2) on the expression of caspase-3 and the changes of ul- tra-structures in protoscolex cells of Echinococcus granulosus in vitro, the protoscolex cells were cultured in vitro with special RMPI-1640 medium in which glutamine was added. Then, 5 mmol/L of H2O2 was added to these cultures for 8 hours to induce cell apoptosis. The condition of apoptosis induced was observed with TUNEL assay and the protein expression of caspase-3 was detected by SP-immunohistochemical technique to stain easpase-3 protein. Meanwhile, the ultra-structure changes in protoscolex cells were observed by means of electron transmission microscopy (TEM). It was found that the expression of caspace- 3 protein and the apoptotic number of protoscolex cells were significantly increased. Under TEM observation, the tero-chromatin in protoscolex cells was also increased and condensation of ehromatin could be demonstrated in some of these cells, appearing as the characteristic features of apoptosis. It is concluded that H2O2 can induce the apoptosis of protoscolex cells of E. granulosus in vitro, in which caspase-3 participates the apoptosis of these cells.
