棘球蚴原头节细胞凋亡的观察

目的观察棘球蚴原头节自身是否存在细胞凋亡现象。方法采集羊肝/肺棘球蚴中自然状态原头节直接用显微镜观察形态结构。过氧化物酶标记链酶卵白素(SP)染色免疫组化法检测半胱天冬氨酸蛋白酶-1(caspase-1)、半胱天冬氨酸蛋白酶-3(caspase-3)表达。原位末端脱氧核糖核苷酸转移酶标记技术(TUNEL法)检测凋亡细胞。透射电镜观察原头节超微结构。结果在自然状态的原头节中,显微镜下见到一些原头节虫体枯萎、结构模糊。这些原头节中的caspase-1、caspase-3呈强阳性表达。TUNEL法检测显示这些原头节中凋亡细胞密布。透射电镜见到典型细胞凋亡形态改变。结论棘球蚴原头节自身存在细胞凋亡现象。

英文摘要:

In order to explore the natural apoptosis in the protoscolex of hydatid cyst,protoscoleces of hydatid cyst from sheep livers or lungs were collected and the morphology was examined directly under microscope. Protein expressions of caspase-1 and caspase-3 were detected by SP immunohistochemical technique. The apoptosis were examined by Terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling assay (TUNEL) and the ultrastructural changes were observed by transmission electron microscope (TEM). Results showed that some withered and dim protoscoleces were found under microscope in natural protoscolex as well as the strong positive expressions of caspase-1 and caspase-3 were found in them. TUNEL assay has detected intensive apoptotic cells and TEM has observed typical ultrastructural changes of apoptosis. It demonstrated that protoscolex of hydatid cyst has apoptotic phenomenon naturally.
