目的:探讨炎症因子Daintain/AIF-1对肝癌细胞耐药性产生的影响。方法:利用MTT法测定耐药HepG2细胞株的IC50,流式细胞术测定耐药细胞株期凋亡率,HPLC方法检测隔耐药细胞株胞内顺铂的外排。结果:Daintain/AIF-1提高了HepG2耐药细胞株的IC50;再次受到相同剂量顺铂的攻击时,Daintain/AIF-1与顺铂联合运用构建的耐药细胞株凋亡率明显下降;Daintain/AIF-1促进了耐药细胞株胞内顺铂的外排。结论:此研究表明Daintain/AIF-1通过影响胞内顺铂的外排而促进了肝癌细胞对顺铂耐药性的产生。
Objective: To investigate the effects of Daintain/AIF- on the production of cisplatin resistance in HepG2 cells. Methods: The IC50 of cisplatin resistant HepG2 cell lines was evaluated by MTT; The apoptosis of cisplatin resistant HepG2 cell lines was detected by flow cytometry analysis; HPLC was performed to detect the efflux of intracellular cisplatin in cisplatin resistant HepG2 cell lines. Results: Daintain/AIF-1 enhanced the IC50 of cisplatin resistant HepG2 cell lines, on the contrary, it decreased the apoptosis when challenged by 10 μg/mL cisplatin again. Further study showed that Daintain/AIF-1 could promote the efflux of intracellular cisplatin in cisplatin resistant HepG2 cell lines. Conclusion: The study indicated that Daintain/AIF-1 enhanced cisplat.in resistance of HepG2 cells by promoting the efflux of intracellular cisplatin.
