目的探讨小鼠心肌梗死时冠状动脉血流动力学变化及其与梗死面积的相关性。方法通过左冠状动脉前降支结扎建立小鼠心肌梗死模型14只,其中左冠状动脉前降支高位结扎7只(高位结扎组),低位结扎7只(低位结扎组);对另7只小鼠进行假手术操作(假手术组)。对3组均行超声生物显微镜(UBM)成像,测量左冠状动脉舒张期峰值血流速度(Vd)、平均血流速度(Vmean)和速度时间积分(VTI);并行组织病理学检查,测量心肌梗死面积(IS)。结果高位结扎组及低位结扎组Vd、Vmean及VTI均较假手术组减低(P均〈0.05),高位结扎组Vmean、VTI均低于低位结扎组(P均〈0.05),高位结扎组与低位结扎组间Vd差异无统计学意义(P〉0.05)。高位结扎组及低位结扎组IS均大于假手术组(P均〈0.05),高位结扎组IS大于低位结扎组(P〈0.05)。小鼠Vd、Vmean及VTI均与IS呈负相关(r=-0.557、-0.693、-0.672,P均〈0.05)。结论小鼠心肌梗死时冠状动脉血流动力学参数减低,且与组织病理学测量的IS呈负相关。
Objective To evaluate the hemodynamic changes in mice models of myocardial infarction and the correlation between hemodynamic parameter and infarct size(IS).Methods Myocardial infarction models of left main coronary artery(LMCA)were established in 14wild-type C57BL/6male mice and divided into low ligation group(L-group)and high ligation group(H-group).Another 7 mice were treated as sham operation group(S-group).Diastolic peak velocity(Vd),mean flow velocity(Vmean)and velocity time integral(VTI)were measured by ultrasound bio-microscopy(UBM).And IS was measured by pathology.Results The Vd,Vmeanand VTI in H-group and L-group were lower those in S-group(all P0.05).And the Vmeanand VTI in H-group were lower than those in L-group(both P0.05).No statistical difference of Vd between H-group and L-group(P〈0.05).The IS in H-group and L-group was separately higher than that in Sgroup(both P〈0.05).While IS in H-group was higher than that in L-group(P〈0.05).The IS was negatively correlated with Vd,Vmeanand VTI of myocardial infarction models in mice(r=-0.557,-0.693,-0.672,all P〈0.05).Conclusion Hemodynamic parameters of coronary artery measured by UBM decrease in mice models of myocardial infarction.And the parameters correlate with IS negatively.
