旨在探讨小花棘豆对大鼠睾丸α-甘露糖苷酶(AMA)的影响,揭示小花棘豆的毒性作用机理。将72只大鼠随机分为4组,即对照组和试验Ⅰ、Ⅱ、Ⅲ组。将小花棘豆全草粉碎后,按Ⅰ组添加150g/kg(含苦马豆素30mg/kg)、Ⅱ组添加300g/kg(含苦马豆素60mg/kg)、Ⅲ组添加450g/kg(含苦马豆素90mg/kg)的比例制作混合饲料,饲喂至典型临床症状出现为止。攻毒后每7d每组随机采集2只大鼠的睾丸,检测大鼠睾丸AMA活性及表达的变化。结果显示,在63d的试验过程中,试验组及对照组大鼠睾丸高尔基体α-甘露糖苷酶Ⅱ(AMA1)和溶酶体α-甘露糖苷酶(AMA2)均有表达,但各试验组表达转录水平与对照组差异显著,试验Ⅰ组大鼠AMA1和AMA2的表达与对照组差异不显著(P〉0.05),但试验Ⅲ组大鼠AMA1和AMA2的表达均显著低于对照,随着试验的进行,抑制效果愈加明显。说明,小花棘豆中毒可影响大鼠睾丸AMA的活性及其基因的转录表达。
The aim of this study is to investigate the effects of Oxytropis glabra DC on content and transcriptional expression ofα-mannosidase(AMA)in testis tissue,and revealed the toxicity mechanism of O.glabra DC.72 rats were divided into 4groups randomly(the control group and experimental groupⅠ,Ⅱ,Ⅲ),The dried plant of O.glabra DC was crushed,and different amounts of the grass powder(150g/kg,300g/kg and 450g/kg,the swainsonine content was 30 mg/kg,60 mg/kg,90mg/kg respectively)were mixed with the feeds in the three experimental groups.The rats consumed the forages freely until typical symptoms were observed.2rats were selected randomly from each group every week,the testicles were collected and the contents of AMA and its mRNA expression in different encephalic regions were measured.In 63 dtest period,We found that AMA in golgi and lysosomal expressed in testicles of all experimental groups,O.glabra DC inhibited the expression of testicles AMA1 and AMA2mRNA,and the difference was obvious,the difference was not obvious in experimental groupⅠ and control group(P〉0.05),the expression of AMA1 and AMA2 mRNA de-creased in experimental group Ⅲ,and it was significantly lower than control group.The results showed that O.glabra DC had remarkable effects on contents and transcriptional expression of AMA in rat testis.
