目的探讨当归及当归多糖对X射线照射致SD大鼠免疫功能损伤的防护作用。方法将40只SD大鼠随机分为对照组、模型组、当归水煎液组、当归多糖组和阳性对照组。常规饲养14d后,各给药组给予相应药物灌胃,对照组和模型组给予等体积蒸馏水灌胃,每日1次,连续7d。第8日除对照组外其余各组大鼠进行全身X射线照射,连续2d,每日1次,每次3Gy,总吸收剂量为6Gy。辐射后第3日股动脉采血处死大鼠,血常规检测外周血白细胞(WBC)、红细胞(RBC)、血红蛋白(HGB)、血小板(PLT),有核细胞计数法观察骨髓有核细胞数量,HE染色观察脾脏病理形态,ELISA检测血清干扰素-γ(IFN-γ)和白细胞介素(IL)-4的含量。结果与对照组比较,模型组大鼠脾脏指数、血WBC、骨髓有核细胞数、血清IL-4和IFN-γ含量明显降低(P<0.05),RBC和HGB明显升高(P<0.05);与模型组比较,各给药组血WBC、骨髓有核细胞数、血清IL-4和IFN-γ含量明显升高(P<0.05),RBC和HGB明显降低(P<0.05)。结论当归及当归多糖对X射线照射致SD大鼠免疫功能损伤具有保护作用。
Objective To investigate the protective effects of Angelicae Sinensis Radix and Angelicae Sinensis Radix polysaccharides on immune function injury induced by X rays in SD rats.Methods Forty SD rats were randomly divided into control group,model group,Angelicae Sinensis Radix Decoction group,Angelicae Sinensis Radix polysaccharide group and positive medicine group.After routine feeding for14day,each administration group was given relevant medicine for gavage,while control group and model group were given the same amount of distilled water for gavage,once a day for7d.From the8th day,except for the control group,the rats in the rest of groups were subjected to whole-body X ray irradiation,continuous exposure to2d;the total absorbed dose was6Gy.The rats were killed by femoral artery after irradiate3d.The WBC count,RBC,HGB,and PLT in peripheral blood were observed by blood routine test;the number of nucleated cells in the bone marrow was observed by nucleated cell count method;the pathological changes of spleen were observed by HE staining under microscope;the contents of IFN-γand IL-4in serum were detected by ELISA.Results Compared with the control group,the spleen index WBC,number of bone marrow nucleated cells and serum contents of IL-4and IFN-γin model group was significantly lower(P<0.05),The contents of RBC and HGB increased(P<0.05).Compared with the model group,WBC,number of bone marrow nucleated cells,and the contents of IL-4and IFN-γin serum of each administration group increased significantly(P<0.05);RBC and HGB decreased significantly(P<0.05).Conclusion Angelicae Sinensis Radix and Angelicae Sinensis Radix polysaccharides have protective effects on the immune function injure induced by X ray in SD rats.
