目的基因的稳定遗传和稳定表达是转基因水稻的必备特性,也是转基因水稻安全评价的重要科目。采用PCR、Southern杂交、RT-PCR、ELISA和除草剂抗性鉴定技术,对转基因抗除草剂水稻Bar68-1进行遗传稳定性分析。研究结果表明,Bar基因已整合在转基因抗除草剂水稻Bar68-1基因组中并可稳定遗传,在转录水平和翻译水平上均可检测到目的基因的稳定表达,在表型上检测出Bar68-1对草铵膦除草剂具有抗性。
Stable inheritance and expression of transgene are essential for genetically modified organisms ( GMO), evaluation of them are im- portant subjects in safety assessment of GMO. The genetic stability of genetically modified herbicide tolerant (GMHT) rice Bar68-1 is as- seased By PCR, Southern blot, RT-PCR, ELISA and glufosinate treatment. The results show that, the Bar gene has integrated in genome of GMHT rice Bin68-1 and inherits stably, the stably expressions of which are detected on the level of transcription and translation respectively, and the glufoeinate resistance of Bar68-1 is verified by glufosinate treatment as well.
