目的:观察针刺血清对体外模拟缺血再灌注后大鼠海马细胞内钙离子的影响,以探讨针灸治疗的体液因素。方法:取新生SD大鼠海马细胞进行原代培养,9~11d后应用荧光分子探针Fluo一3AM染色,激光共聚焦扫描显微镜动态扫描观察加入正常大鼠血清和电针“百会”“足三里”“曲池”“三阴交”2周后的血清对海马细胞内钙离子的影响。结果:加入正常血清,钙离子荧光值由461±96变为697±113;加入针刺血清,钙离子荧光值由673±108变为584±103。说明加入针刺血清后,可以降低升高的细胞内钙离子的浓度。结论:针刺血清中存在着某种活性物质可以明显地降低缺血再灌注后细胞内钙离子浓度,从而为针灸治疗疾病作用中体液因素的存在提供了直接依据。
Objective To investigate the effect of acupuncture serum on Ca^2+ content in the cultured nervous cells of hippocampus after ischemia-reperfusion, so as to probe into humoral factors in acupuncture treatment. Methods The neurons of the hippocampus from the new born rats were cultured for 9--11 days. Fluorescein-molecular probe Fluo-3 AM was used for staining of intraeellular Ca^2+. Fluorescent levels in the nervous cells cultured with the ser- um of the normal rats or the rats given electroacupuneture at "Baihui" (GV 20), "Zusanli" (ST 36), "Quchi" (LI 11) and "Sanyinjiao" (SP 6) for 2 weeks were determined by using a laser confocal microscope. Results After the normal serum was added, the intracellular Ca^2+ fluorescent levels increased to 697±113 from 461 ± 96, while after acupuncture serum was added, the Ca2. fluorescent levels decreased to 584+ 103 from 673±108, indicating that after addition of acupuncture serum, the increased intracellular Ca^2+ content could be decreased. Conclusion There are some active substances in acupuncture serum which can obviously decrease intracellular Ca^2+ content'after ischemia-reperfusion, so as to provide a direct evidence for role of humoral factor in acupuncture treatment.
