目的:通过采用D-273内质网荧光分子探针来观察针刺血清调节内质网活性的作用,从而探讨针刺作用的细胞分子水平机理。方法:选用原代培养的9-11天的新生SD大鼠海马细胞,应用激光共聚焦显微镜动态扫描观察加入正常大鼠血清和电针“百会”、“足三里”、“曲池”、“三阴交”2周后的血清对培养的正常和模拟缺血缺氧海马神经元内质网活性的影响。结果:加入正常血清后对照和模型组海马神经元内质网荧光值均下降,而加入针刺血清后下降的荧光强度变化值减小。结论:针刺血清阻抑了神经元内质网活性的下降,显示针刺血清可能有提高海马神经元的内质网活性效应。
Objective:To observe the effect of acupuncture serum on endoplasmic reticulum activity of hippocampal neurons ischemic reperfusion injury by using the D-273 as fluorescent molecular probes.Methods:Applying the normal and ischemia refutation model in vitro,the changes of endoplasmic reticulum fluorescence in the hippocampus were observed by using laser confocal scanning microscope and staing D-273.Results:After adding normal serum,endoplasmic reticulum fluorescence decreased on control and model group hippocampal neurons Compaired with control serum,we found the level of decreasing intensity of the fluorescence was added down after acupuncture serum significantly.Conclusion:Acupuncture serum may increase the endoplasmic reticulum Activities in the hippocampal neurons.
