目的:探讨MACC1基因与肾癌转移的相关性,及其表达降低对肾癌细胞侵袭能力的影响。方法:应用荧光定量PCR检测原发未转移肾癌和原发伴转移肾癌组织中MACC1 mRNA的表达。设计并合成MACC1基因特异性的siRNA,转染肾癌Caki-1细胞,检测转染后MACC1蛋白的表达以及Caki-1细胞的侵袭能力。结果:原发伴转移肾癌组织中的MACC1 mRNA表达较原发未转移肾癌明显增加(P〈0.05)。siRNA-MACC1能够明显抑制Caki-1细胞中MACC1蛋白的表达(P〈0.05)。MACC1蛋白表达抑制的Caki-1细胞的侵袭能力明显降低(P〈0.05)。结论:MACC1基因与肾癌的转移相关,其表达抑制能够降低Caki-1细胞的侵袭能力。
Objective:To research the relationship between MACC1 gene and renal carcinoma and the influence of MACC1 expression change to invasiveness of Caki-1 cells.Methods:Fluorescent quantitative PCR was used to detect MACC1 mRNA in primary renal carcinoma without metastasis and primary renal carcinoma with metastasis.Specific siRNA for MACC1 gene was designed and synthesized,and then Caki-1 cells were transfected with siRNA-MACC1 to inhibit the expression of MACC1 gene.After RNA interference,invasiveness of Caki-1 cells was detected.Results:The expression of MACC1 mRNA in primary renal carcinoma with metastasis was much higher than that in primary renal carcinoma without metastasis(P0.05).The expression of MACC1 protein in Caki-1 cells transfected with specific siRNA decreased significantly(P0.05).After RNA interference,the invasiveness of Caki-1 cells reduced significantly(P0.05).Conclusion:There is a relationship between MACC1 gene with metastasis of renal carcinoma,and the inhibition of MACC1 gene expression can inhibit the invasiveness of Caki-1 cells.
