目的研究MTSS1基因在膀胱癌中组织的表达及其对膀胱癌EJ138细胞增殖和凋亡的影响。方法荧光定量PCR检测41例膀胱癌及相应癌旁正常组织中MTSS1基因的表达。将MTSS1基因特异性的siRNA转染EJ138细胞。转染后,Western Blot检测转染后MTSS1蛋白的表达,MTT法测定细胞生长抑制率,流式细胞法检测细胞凋亡率。结果与癌旁正常组织相比,MTSS1 mRNA在膀胱癌组织中表达明显下调。RNA干扰能够显著抑制EJ138细胞中MTSS1蛋白的表达;MTSS1基因表达降低后,EJ138细胞的生长抑制率明显降低,细胞增殖活力明显增加,而且凋亡率明显降低。结论 MTSS1基因在膀胱癌低表达,促进膀胱癌细胞增殖并抑制细胞凋亡。
Objective To study the expression of MTSS1 gene in bladder carcinoma and the influence of its low expression to proliferation and apoptosis in EJ138 cells.Methods Real-time PCR was used to detect the expression of MTSS1 gene in 41 cases of bladder carcinoma and peri-cancerous normal tissues.RNA interference was used to inhibit the expression of MTSS1 gene to study the expression of MTSS1 gene by western blot,the inhibition ratio of cell proliferation was detected in transfected cells by methyl thiazolyl tetrazolium(MTT) and the apoptosis rate was detected by flowcytometry.Results The expression of MTSS1 gene presented an obvious decreasing in bladder carcinoma compared to peri-cancerous normal tissues.After RNA interference,the expression of MTSS1 protein in the transfected EJ138 cells was downregulated significantly,the inhibitory rate of cell proliferation decreased significantly,the proliferation increased significantly,and the apoptosis rate decreased significantly in transfected cells.Conclusions The downexpression of MTSS1 gene in bladder carcinoma is related to the premotion of EJ138 cells proliferation and inhibition of EJ138 cells apoptosis.
