目的探讨了NF-kβ在KISS-1调控MMP-9的表达中的作用。方法设计并合成了KISS-1基因特异性的siRNA,转染肾癌caki-1细胞,westernblot检测KISS-1、NF-KB和MMP-9蛋白的表达。应用NF-kβ抑制剂BAY11-7082处理KISS-1基因表达沉默的caki-1细胞,westernblot检测MMP-9蛋白的表达。结果在Caki-l细胞中,KISS-1基因表达沉默能够显著上调NF-kβ蛋白的表达,并显著下调MMP-9蛋白的表达。而BAY11-7082能够在KISS-1基因表达沉默的Caki-1细胞,显著上调MMP-9蛋白的表达。结论KISS-1基因在肾癌细胞中能够通过NF-kβ调控MMP-9的表达。
[ Objective ] To research the effects of NF - KB on KISS-1 regulate the expression of MMP-9 in re- nal carcinoma. [ Methods] Specific siRNA for KISS-1 gene was designed and synthesized, and then Caki-1 cells were transfected with siRNA - KISS-1 to inhibit the expression of KISS-1 gene. After RNA interference, the ex- pression of KISS-1, NF -kβ and MMP - 9 were detected by western blot. BAY 11 - 7082 was used to treat the Ca- ki - 1 cells with KISS - 1 silence, the expression of MMP - 9 were detected by western blot. [ Results ] In Caki - 1 cells, KISS - 1 silence could up - regulate the expression of NF - kβ, and down - regulate the expression of MMP - 9 remarkably. After BAY 11 - 7082 was used to treat the Caki - 1 cells with KISS - 1 silence, the expression of MMP - 9 up - regulated remarkably. [ Conclusion] KISS - 1 gene can regulate the expression of MMP - 9 by NF -kβ in renal carcinoma cells.
