为了研究副鸡禽杆菌的荚膜多糖的构成及生物学活性,本试验提取了B型副鸡禽杆菌的荚膜多糖,测定其含量,并分析单糖组分及分子量大小。用乙醇沉淀法在细菌培养液上清中提取了荚膜多糖,利用25%乙醇去核酸、苯酚抽提去蛋白,最后以苯酚-硫酸法测定其含量,高效液相色谱(HPLC)分析其组分及分子量大小。结果显示,每升菌液可以抽提得到荚膜多糖85mg,含量为59.6%,该多糖主要由L-鼠李糖、D-木糖、D-葡萄糖等单糖组成,总分子量约209Ku。
In order to research the composition and biological activities of capsular polysaccharide(CPS)from Avibacterium paragallinarum(Apg),Apg CPS was extracted,and followed by the detection of its contents and analysis of constituents and molecular weight. Ethanol precipitation was used to extract CPS from supernatant,25% ethanol and phenol were used to dislodge nucleic acid and protein respectively, phenol-vitriolic was used to detect its contents,high performance liquid chromatography(HPLC)was used to analyze its composition and molecular weight. Results show that about 85 mg CPS is gained from a liter bacteria liquid, which is composed of L-rhamnose D-xylose D-glucose and an unknown monosaccharide. Its purity is 59.6% and the molecular weight is about 209 Ku. It lays the foundation for further study of the biological function of avibacterium paragallinarum capsules.
