目的:观察夏枯草总三萜(TTP)对血小板衍生因子(PDGF-BB)干预的肝星状细胞(HSCT6)中TGF-β1/Smad通路调控以及其抑制HSC增殖的作用。方法:用不同浓度的TTP(10、30、100、300、1 000μg/mL)及PI3K抑制剂(LY294002,10μmol/L)对PDGF-BB刺激的HSC-T6进行处理,四甲基偶氮唑蓝(MTT)法检测细胞增殖;RT-PCR和Western blot法检测HSC-T6中TGF-β1信号转导通路下游中介分子smad2、smad3、smad7表达。结果:TTP给药组可抑制PDGF诱导的HSC-T6增殖,下调smad2、smad3表达,升高smad7表达。结论:TTP对PDGF-BB刺激的HSC-T6增殖具有较好的抑制作用,其机制可能与阻断TGF-β1/Smad信号通路传导有关。
AIM: To investigate the inhibitory role of total triterpenoids of prunella vulgaris L.( TTP) in the proliferation,and regulatory effects on the TGF-β1/ Smad pathway in hepatic stellate cellT6( HSC-T6) stimulated by PDGF. METHODS:HSC-T6 cells were stimulated by PDGF-BB and treated with different does of TTP( 10,30,100,300, 1000 μg / m L) and PI3 K inhibitor( LY294002,10 μmol/L),the HSC-T6 proliferation was detected by MTT,the expression of mRNA and protein levels of smad2,smad3,smad7 were detected by RT-PCR and Western blot. RESULTS:As compared with the model group,TTP could inhibit HSC-T6 stimulated by PDGF-BB. Furthermore,TTP reduced the mRNA and protein expression of smad2,smad3 and increased smad7. CONCLUSION: TTP can inhibit the proliferation of HSC stimulated by PDGF-BB, the mechanism of this process may be related to the interference with TGF-β1/ Smad signal transduction pathway.
