目的 研究端粒调控因子Tankyrase和端粒酶逆转录酶(hTERT)在胃腺癌组织中的表达水平。并进行相关分析。方法应用荧光实时定量PCR(Real-time PCR)技术检测16例胃腙癌组织和离肿瘤边缘5cm处正常癌旁组织中的Tankyrase和hTERT mRNA表达水平,同时分析Tankyrase表达水平在不同胃腺癌临床分期和组织分化程度各组间的关系。结果胃腺癌中Tankyrase mRNA的表达(1.44×10^2,3.88×10^-5~0.4847)高于正常对照(1.0134×10^-2,0~4.933×10^-2)(T=31,P〈0.05)。hTERT mRNA在胃腺癌中的表达为(1.235×10^-4~0.13301),显著高于正常对照(0,0~4.38×10^-5)(T=0,P〈0.01),两者呈正相关(r=0.553,P〈0.05)。但在不同胃腺癌临床分期、组织分化程度各组之间差异均无统计学意义。结论Tankyrase和hTERT在胃腺癌中表达增高,呈正相关,可能是胃腺癌发生过程中的重要因素。
Objective To detect the expression level of telomere regulating factor Tankyrase and human telomerase reverse transcriptase(hTERT) in gastric adenocarcinoma and further elucidate its correlation. Methods Quantitative real-time PCR was used to measure expression of Tankyrase and hTERT mRNA in canerous and normal adjacent tissues from 16 patients with gastric adenocarcinoma. Results The expression of Tankyrase mRNA in gastric adenocarcinomas(1.44×10^2,3.88×10^-5~0.4847) was significantly higher than that in normal adjacent mueosas( 1.0134×10^-2,0~4.933×10^-2 ) ( p 〈0.05). The expression of hTERT mRNA in gastric adenocarcinomas( 1.235×10^-4~0.13301 )was also significantly higher than that in normal adjacent mucosas(0,0~4.38×10^-5 ) ( P 〈 0.01 ). The expression of Tankyrase correlated positively with hTERT(Spearman correlation was 0.553, P 〈 0.05). But no significant differences were seen between different clinical stages and different pathological grades. Conclusion Tankyrase is overexpressed in gastric adenocareinoma and correlated positively with hTERT. It may be one of the important factors in gastric adenocarcinomas.
